This is not the place to make this change. CIRCUITPY_USB_HOST should already be turned on for this board, in ports/raspberrypi/mpconfigboard.mk. Use the 10.0.0-alpha.7 or later release if 9.2.x doesn't have it.
EDIT: ... and I looked, it's turned on in both 9.2.x and 10.0.0 builds. See https://docs.circuitpython.org/en/latest/shared-bindings/support_matrix.html
